Description

ASCH AST-01 Skin Stratum Corneum Stripping Tape

Application

Stratum Corneum Stripping In ex vivo skin drug tissue distribution experiments, the stratum corneum is stripped and removed to evaluate the penetration depth of drugs in the stratum corneum. Skin Barrier Function Evaluation The integrity and function of the skin barrier can be assessed by analysing the number of stratum corneum cells collected on the tape. Non-invasive Skin Test By application and stripping, SC Checker tape can non-invasively remove stratum corneum cells from the skin surface. Efficacy Evaluation of Cosmetics and Skincare Products It is used to assess the effects of cosmetics and skincare products on skin barrier function, such as exfoliation, moisturising effect and skin softness.

Product Description
The therapeutic efficacy of topical dermatological products depends on the rate and extent at which active ingredients reach the target sites within the skin, namely the stratum corneum (SC), viable epidermis, or deeper dermis. The tape stripping (TS) method has been extensively investigated for many years as a technique to estimate in vivo dermatopharmacokinetics (DPK). TS employs adhesive tapes to collect successive layers of the stratum corneum (SC) after topical application of formulations, followed by quantification of drug content in each stripped skin layer. Combined with analytical techniques, TS is used to measure the penetration of chemicals into the SC over time and evaluate the performance of topical drug formulations.
According to the Technical Guideline for In Vitro Release Test (IVRT) and In Vitro Permeation Test (IVPT) of Locally Acting Chemical Generic Drugs (Draft for Comment) issued by CDE in July 2024, "Depending on the research objectives, additional assessments of drug tissue distribution (e.g., drug amounts in the stratum corneum, viable epidermis and dermis) and overall drug mass balance may be required". It is generally necessary to completely strip the stratum corneum from excised skin using adhesive tapes.
ASCH AST-01 Skin Stratum Corneum Tape Stripping Procedure:
(a) Apply the tape over excised pig skin sections with a diameter of 25 mm.
(b) Place a 2 kg standard weight on the tape surface for approximately 10 seconds to compress the tape and skin section.
(c) Rapidly strip the tape from the skin using forceps or gloved fingers; perform about 20 stripping cycles (a new tape is used for each cycle).
(d) Use the Labodorf SquameScan to measure infrared absorbance to determine the amount of residual stratum corneum on the tape. The stripping endpoint is confirmed when the absorbance value approximates that of blank tape.
